Background
Epinephrine (also known as adrenaline) is a hormone and a neurotransmitter. It increases heart rate, constricts blood vessels, dilates air passages and participates in the fight-or-flight response of the sympathetic nervous system. Chemically, epinephrine is a catecholamine, a monoamine produced only by the adrenal glands from the amino acids phenylalanine and tyrosine. Norepinephrine or noradrenaline is a catecholamine with multiple roles including as a hormone and a neurotransmitter. Areas of the body that produce or are affected by norepinephrine are described as noradrenergic. Norepinephrine is synthesized from dopamine by dopamine beta-hydroxylase. It is released from the adrenal medulla into the blood as a hormone, and is also a neurotransmitter in the central nervous system and sympathetic nervous system where it is released from noradrenergic neurons in the locus coeruleus.

Assay Information
Adrenaline (epinephrine), noradrenaline (norepinephrine), and dopamine are extracted by using a cisdiolspecific affinity gel, acylated and then converted enzymatically. The assay procedure follows the basic principle of radioimmunoassay, involving competition between a radioactive and a nonradioactive antigen for a fixed number of antibody binding sites. The amount of 125Ilabelled antigen bound to the antibody is inversely proportional to the analyte concentration of the sample. When the system is in equilibrium, the antibody bound radioactivity is precipitated with a second antibody in the presence of polyethylene glycol. The precipitate is counted in a gamma counter. Determination of unknown samples is achieved by comparing their activity with a reference curve prepared with known standards.
