Background |
Myasthenia Gravis (MG) is a skeletal muscle disorder characterized by muscular weakness. A good correlation is observed between antiacetylcholine receptor (AChR) antibodies and muscle weakness in individual patients . Anti AChR antibodies are present in approximately 90% of patients with MG. Anti AChR antibodies could be: binding antibodies (multitudes of wide populations of antibodies directed to hydrophilic domains of receptors), blocking antibodies (preventing binding of acetylcholine to receptors) and modulating antibodies (accelerating endocytosis resulting in loss of receptors). |

Acetylcholine receptor (AChR) modulating antibodies eliminate binding sites for the cholinergic antagonist, 125Ialphabugarotoxin by a) direct steric or allotter inhibition, or b) indication of detected via a radiobinding assay. Living cultures of a human skeletal muscle cell line are incubated overnight with patient’s serum. 125Ialpha bungarotoxin is added and allowed to complex with the AChR on the membranes. Binding specificity is determined by separation of the free 125Ialphabugarotoxin by repeated washing .The bound fraction is then measured on a gamma counter. |
