The enzyme-linked immunosorbent assay (ELISA) is a test that uses antibodies and an observed change in color to identify the presence of a target of interest. The purpose of an ELISA is to determine if a particular protein is present in a sample and how much is present, ie ELISAs can be both quantitive and qualitive.
In an ELISA assay, a target antigen is immobilized to a solid surface and then complexed with an antibody that is linked to an enzyme. Detection is accomplished by assessing the conjugated enzyme activity via incubation with a substrate which may or may not contain the target to produce a measureable product. When the enzyme reaction has incubated, the plate is placed into a plate reader and the amount of colored product is determined for each well. The amount of color produced is proportional to the amount of primary antibody bound to the proteins on the bottom of the wells.
ELISA is a flexible assay and can be performed with a number of useful modifications to the basic procedure. These include Direct, Indirect, Competitive and ELISPOT (enzyme-linked immunospot assay) which captures target proteins in a similar fashion to a Western Blot.
