Flow Cytometric assay using BCAS3 antibody (10R-1295)

HeLa cells were fixed in 2% paraformaldehyde/PBS and then permeabilized in 90% methanol. Cells were stained with anti-BCAS3 antibody (shaded) or isotype control (unshaded) followed by Alexa Fluor 488 conjugated goat anti-mouse IgG.

Method of Purification BCAS3 antibody was purified using protein G column chromatography from culture supernatant of hybridomacultured in a medium containing bovine IgG-depleted(approximately 95%) fetal bovine serum.

General Information

Biological Significance Breast carcinoma amplified sequence 3, also known as BCAS3, is a protein which in humans is encoded by the BCAS3 gene. BCAS3 is a gene that is amplified and overexpressed in breast cancer cells. The BCAS3 gene is regulated by estrogen receptor alpha. The PELP1 protein acts as a transcriptional coactivator of estrogen receptor induced BCAS3 gene expression. In addition BCAS3 possesses histone acetyltransferase activity and itself appears to act as a coactivator of ER alpha. Furthermore BCAS3 requires PELP1 to function as a coactivator in ER alpha. Hence BCAS3 apparently is involved in a positive feedback loop leading to ER alphamediated signal amplification.
