Casein is a milk-derived substance routinely used in blocking buffers for Western, Southern and Northern Blot procedures. Blocking agents are an important element in reducing signal noise in a blotting procedure as well as to reduce non-specific binding of detection reagents to the membranes  and microplates in antibody-based detection procedures. Casein buffer is also commonly used to block background binding of antibodies in ELISA assays as well as for use in Immunohistochemistry and nucleic acid detection methods.
