General Information

Product Use Monoclonal CD71 antibody specific for Human CD71 for use in Flow Cytometry.
Biological Significance CD71 is the transferrin receptor. CD71 regulates the cellular uptake of iron occurs via receptor-mediated endocytosis of ligand-occupied receptors into specialized endosomes. Endosomal acidification leads to iron release. The apotransferrin-receptor complex is then recycled to the cell surface with a return to neutral pH and the concomitant loss of affinity of apotransferrin for its receptor. Transferrin receptor is necessary for development of erythrocytes and the nervous system.
Expression CD71 is expressed in a wide range of cell types and tissues. Expression level is highest in lymphocytes, placenta and neoplastic cells. CD71 is a single-pass type II membrane protein localized to the cell membrane.
Implications in Disease Overexpression of CD71in malignant cells mediates higher iron uptake required for cell division. Expression is activated by c-Myc. No mutation or variation in CD71 causes cancer and CD71 is not involved in cancer-associated translocations. CD71 variant S142G modifies the associations of HFE C282Y mutation in cancer susceptibility for hepatocellular carcinoma, breast cancer, leukemia, colorectal cancer and multiple myeloma. Biological plausibility of these associations has been supported by the successful use of monoclonal antibodies against transferrin receptor in cancer treatment in vitro and in vivo. There are no disease-causing mutations in the CD71 gene. However, there are missense coding region variants that may have functional effects. The only one with appreciable frequency (rs3817672) is in exon 4 and encodes S142G amino acid substitution. This polymorphism does not have a homogeneous global distribution. Its minor allele in Caucasians is the major allele in Asians and Africans.
Sequence Information CD71 is composed of 760 amino acids with a molecular weight of 84871 Da.
Post-Translational Modifications D71 is N- and O-glycosylated, phosphorylated and palmitoylated. The serum form is only glycosylated CD71 is proteolytically cleaved on Arg-100 to produce the soluble serum form (sTfR) and is palmitoylated on both Cys-62 and Cys-67. Cys-62 seems to be the major site of palmitoylation. There is also ubiquitination at Lys39, Lys224, Lys344, Lys358, Lys720 and glycosylation at Thr104, Asn251, Asn317, Asn727.
