Flow Cytometric assay using EIF2S1 antibody (10R-1275)

HeLa cells were fixed in 2% paraformaldehyde/PBS and then permeabilized in 90% methanol. Cells were stained with anti-EIF2S1 antibody (shaded) or isotype control (unshaded) followed by Alexa Fluor 488 conjugated goat anti-mouse IgG.

Method of Purification EIF2S1 antibody was purified using protein G column chromatography from culture supernatant of hybridoma cultured in a medium containing bovine IgG-depleted (approximately 95%) fetal bovine serum.

General Information

Biological Significance The translation initiation factor eIF2 catalyzes the first regulated step of protein synthesis initiation, promoting the binding of the initiator tRNA to 40S ribosomal subunits. Binding occurs as a ternary complex of methionyl-tRNA, eIF2, and GTP. eIF2 is composed of 3 nonidentical subunits, alpha (36 kDa), beta (38 kDa, MIM 603908), and gamma (52 kDa, MIM 300161). The rate of formation of the ternary complex is modulated by the phosphorylation state of eIF2-alpha.
