General Information

Product Use Monoclonal ICAM1 antibody specific for Human ICAM1 for use in ELISA and Western Blot.
Biological Significance ICAM1 is a member of the immunoglobulin superfamily, the superfamily of proteins including antibodies and T-cell receptors. ICAM1 is a transmembrane protein possessing an amino-terminus extracellular domain, a single transmembrane domain, and a carboxy-terminus cytoplasmic domain. ICAM1 is known for its importance in stabilizing cell-cell interactions and facilitating leukocyte endothelial transmigration. More recently, ICAM1 has been characterized as a site for the cellular entry of human rhinovirus. Because of these associations with immune responses, it has been hypothesized that ICAM1 could function in signal transduction. ICAM1 also plays a major role in spermatogenesis due to its antagonistic effects on the tight junctions forming the blood-testis barrier.
Expression ICAM1 is typically expressed on endothelial cells and cells of the immune system. ICAM1 is a type of intercellular adhesion molecule continuously present in low concentrations in the membranes of leukocytes and endothelial cells. Upon cytokine stimulation, the concentrations greatly increase. ICAM1 is also located in the vascular endothelium, macrophages, and lymphocytes.
Implications in Disease ICAM1 has been implicated in subarachnoid hemorrhage (SAH). Levels of ICAM1 are shown to be significantly elevated in patients with SAH over control subjects in many studies. ICAM1 expressed by respiratory epithelial cells is also the binding site for rhinovirus, the causative agent of most common colds. ICAM1 has an important role in ocular allergies recruiting pro-inflammatory lymphocytes and mast cells promoting a type I hypersensitivity reaction.
Sequence Information ICAM1 is composed of 532 amino acids with a molecular weight of 57825 Da. It contains 5 Ig-like C2-type (immunoglobulin-like) domains at positions 41 – 103, 128 – 193, 230 – 297, 325 – 378 and 412 – 464. There is a cell attachment motif at positions 152 – 154.
Post-Translational Modifications ICAM1 is monoubiquitinated, which is promoted by MARCH9 and leads to endocytosis. Glycosylation occurs at Ile60, Asn130, Asn145, Asn183, Asn202, Asn267, Asn296, Asn385 and Asn406.
