Biotin is a water soluble B vitamin necessary for cell growth, the production of fatty acids, and the metabolism of fats and amino acids. Biotin is widely used throughout the biotechnology industry conjugated to proteins and antibodies as a means of tagging a target molecule for biochemical analysis and other procedures such as purification. Biotins small size means the biological activity of the conjugated will be unaffected.
Avidin, found in raw egg white, is a highly cationic glycoprotein with an extremely high affinity for biotin. Streptavidin is a nonglycosylated protein isolated from Streptomyces avidinii. Streptavidin-biotin is one of the strongest non-covalent interactions in nature. In addition to its use in purification, thanks to the strong streptavidin-biotin bond, steptavidin is also used to attach biomolecules to solid supports such as microtitre wells.
Streptavidin and Avidin-biotin systems have been established as extremely powerful tools in biological sciences and used in a large range of biochemical assays, including Western Blot, ELISA, Immunohistochemistry, Immunofluorescence and ELISPOT to amplify signals.
Antibodies labeled with biotin greatly increase the sensitivity of an assay by its ability to amplify a given reaction. A biotin/streptavidin (avidin) system offers significant sensitivity and low background noise. Due to its high affinity bond, it minimizes nonspecific interactions with other proteins, and permits more complete washing without the risk of lowered signal due to antibody detachment. This results in significantly enhanced sensitivity and the ability to detect minute amounts of antigens.
Fitzgerald Industries offers a huge range of biotinylated secondary antibodies. We supply biotinylated versions of all the major immunoglobulin classes, subclasses and fragments and expressed in all the commonly utilized host species including rabbit, goat, donkey, sheep, chicken, mice, pigs and more, specific for human and animal immunoglobulins.
Some of our top selling biotinylated secondary antibodies for use in biotin/streptavidin detection systems include:
Rabbit anti Sheep IgG (biotin) was purified by affinity chromatography and is recommended for use in ELISA, Immunohistochemistry and Western blot.
Rabbit anti Human IgA (biotin) is also very popular due to its suitability for several kinds of immunoassay.
Rabbit anti Mouse IgG1 (biotin) is one our many biotinylated immunoglobulin subclasses, this one is also recommended for use in ELISA, Immunohistochemistry and Western blot.
Donkey anti Rabbit IgG (H + L) (Fab'2) (biotin) is an excellent example of our range of biotinylated Fab fragment antibodies; this one is suitable for multiple applications including Flow Cytometry.
